Comment Both caffeine and 2,4-dinitrophenol (DNP) significantly induce PGC-1alpha, and increase both metabolism and mitochondrial content in skeletal muscle. Caffeine increases metabolism through binding of phosphodiesterase resulting in increased cyclic adenosine monophosphate (cAMP). Increasing cAMP activates AMP-activated protein kinase (AMPK), protein kinase A, and cAMP response element-binding causing the transcription of genes associated with increased fat oxidation.
