Comment | The effect of macrophage-specific PPAR-gamma knockout on insulin signaling in skeletal muscle and liver was a significant reduction in insulin-stimulated tyrosine phosphorylation of insulin receptor substrate 1 (IRS-1) along with a corresponding decrease in serine phosphorylation of Akt in both tissues. Reductions in IRS-1 and Akt phosphorylation in muscle and liver were paralleled by significant increases in the activation of IKK-alpha/beta (phospho-IKK) and JNK (phospho-JNK), serine kinases previously shown to cause inhibition of IRS-1 function and insulin signaling. |
