Four hours after treatment, PTP1B mRNA levels were increased by 4.5-fold in liver, 1.8-fold in skeletal muscle, 1.6-fold in adipose tissue, and 1.4-fold in the arcuate nucleus. To determine whether TNF-alpha is necessary for PTP1B overexpression in insulin- and leptin-target tissues in mice with diet-induced obesity, the authors compared the level of PTP1B overexpression in TNF-alpha-deficient (TNF-alpha -/-) and wild-type (TNF-alpha +/+) mice with diet-induced obesity. PTP1B overexpression in TNF-alpha -/- mice was significantly lower in both female and male mice compared with TNF-alpha +/+ mice.
